Visualisation of proteome-wide ordered protein abundances in Trypanosoma brucei

چکیده
Background: Trypanosoma brucei is a protozoan parasite and etiological agent of human animal African trypanosomiasis. It has complex life cycle, but the most studied cellular types are in vitro cultivated bloodstream- procyclic-forms. These correspond to replicating, mammalian host bloodstream-dwelling, slender trypomastigotes tsetse vector midgut-dwelling procyclic lifecycle stages, respectively. Several proteomics studies have reported differential abundance proteins between these vitro cultivated cell types. However, there no datasets providing protein abundance, from least abundant, within both types. Methods: We used MaxQuant software 1.6.10.4 reprocess recent large-scale experiment our laboratory extracted intensity-based quantifications bloodstream form proteomes. Results: created web interface visually explore abundances T. brucei Conclusions: The visualization tool, searchable by name(s) attribute(s), is likely be useful trypanosome research community. will allow users contextualise their interest in terms proteomes.

The nuclear proteome of Trypanosoma brucei
Trypanosoma brucei is a protozoan flagellate that is transmitted by tsetse flies into the mammalian bloodstream. The parasite has a huge impact on human health both directly by causing African sleeping sickness and indirectly, by infecting domestic cattle. The biology of trypanosomes involves some highly unusual, nuclear-localised processes. These include polycistronic transcription without cla...
متن کاملProtein isoprenylation in Trypanosoma brucei brucei.
Trypanosoma brucei is a digenetic protozoan parasite presenting a public health hazard in certain areas of Africa; it is transmitted between mammals (bloodstream form (BSF)) by tsetse flies (procyclic form (PCF)). We have an ongoing interest in small GTP proteins in T. brucei, and these are commonly isoprenylated. Many isoprenylated proteins belong to the rab family and include ras; the isopren...
متن کاملOrdered assembly of the duplicating Golgi in Trypanosoma brucei.
The new Golgi in the protozoan parasite Trypanosoma brucei grows near to the old and adjacent to the growing new endoplasmic reticulum exit site. Growth is now shown to be at least a two-stage process, in which a representative matrix marker (GRASP) and enzyme (GntB) are delivered to the site of assembly, followed approximately 10 min later by a COPI component (epsilon-COP) and a trans-Golgi ne...
متن کاملA Protein Complex Map of Trypanosoma brucei
The functions of the majority of trypanosomatid-specific proteins are unknown, hindering our understanding of the biology and pathogenesis of Trypanosomatida. While protein-protein interactions are highly informative about protein function, a global map of protein interactions and complexes is still lacking for these important human parasites. Here, benefiting from in-depth biochemical fraction...
متن کاملTrypanosoma brucei brucei
Protein synthesis in Trypanosoma brucei brucei was rapidly inhibited during polyamine depletion by DL-adifluoromethylornithine (DFMO) in vitro and in vivo. [3H]Leucine incorporation was depressed 30-40 % by 24 h and 80-90 % by 48 h of DFMO treatment. Concomitantly there was an apparent decrease in the synthesis of the variant-specific glycoprotein (VSG) in DFMO-treated trypanosomes, as measured...
